摘要:
目的:运用网络药理学探析加味射干麻黄汤治疗急性支气管炎的作用机制。方法:首先,运用中药系统药理学数据库(TCMSP)获取加味射干麻黄汤的有效成分及其作用靶点,在GeneCards数据库中以“acute bronchitis”为关键词,寻找急性支气管炎的疾病相关靶点,再将有效成分作用靶点与疾病靶点相互取交集,得到加味射干麻黄汤治疗急性支气管炎的基因靶点;其次,利用STRING数据库构建蛋白互作(PPI)网络,根据度值计算网络核心靶点;最后,采用R软件进行GO富集和KEGG通路分析。结果:从TCMSP数据库获得加味射干麻黄汤有效活性成分157个,急性支气管炎疾病靶点1 083个,相互取交集得出加味射干麻黄汤治疗急性支气管炎的潜在靶点共93个,GO功能富集分析得到生物过程1 166条,分子功能44条,细胞成分27条。KEGG富集分析得到157条条目。结论:加味射干麻黄汤可能通过槲皮素调节白细胞介素-6(IL-6)细胞因子来保护气道屏障及抗炎等途径来有效治疗急性支气管炎。
Abstract:
bjective: To explore the mechanism of action of Jiawei Shegan Mahuang decoction in the treatment of acute bronchitis by using network pharmacology. Methods: First of all, he effective components and their action targets of Jiawei Shegan Mahuang decoction and their action targets were obtained by using the Traditional Chinese Medicine System Pharmacology Database (TCMSP), and the disease related targets of acute bronchitis were found by using the keyword "acute bronchitis" in the GeneCards database, and then, the effective component action targets and disease targets were intersected to obtain the gene targets of Jiawei Shegan Mahuang decoction for the treatment of acute bronchitis. Then, the protein interaction (PPI) network was constructed by using STRING database, and the core target of the network was calculated according to the degree value. Finally, the GO enrichment and KEGG pathway analysis were carried out by using R software. Results: From the TCMSP database, 157 effective active components of Jiawei Shegan Mahuang decoction and 1083 disease targets of acute bronchitis were obtained, and 93 potential targets of Jiawei Shegan Mahuang decoction for treating acute bronchitis were obtained by mutual intersection. 1166 biological processes, 44 molecular functions and 27 cell components were obtained by GO function enrichment analysis. 157 samples were obtained by KEGG enrichment analysis. Conclusion: Jiawei Shegan Mahuang decoction may effectively treat acute bronchitis through quercetin regulating interleukin-6 (IL-6) cytokines to protect the airway barrier and anti-inflammatory.
