摘要:
目的：建立炎见宁片中毛冬青活性成分冬青素A的高效液相色谱含量测定方法，考察不同批次炎见宁中冬青素A的含量，为炎见宁片的质量控制提供方法学依据。方法：采用Supersil ODS2 C18（4.6 mm × 200 mm，5 μm）色谱柱；流动相乙腈(A)-0.1%磷酸(B)，梯度洗脱；柱温25℃；流速1 mL·min-1；检测波长210 nm，进样量20 μL，利用高效液相色谱分析法（High performance liquid chromatography，HPLC）测定炎见宁片中冬青素A的含量，分别进行专属性、精密度、稳定性、重复性、检出限、定量限、加样回收率等考察以验证方法的科学性。结果：冬青素A在浓度2.7～108 μg·mL-1范围内线性良好（r≥0.9999）；专属性实验表明炎见宁片中冬青素A具有专属性；精密度为RSD=1.00%；稳定性为RSD=4.24%；重复性RSD=0.77%；平均加样回收率为90.3%，RSD=7.3%（n=9）。结论：该方法灵敏、准确、可靠且分离度好，可用于炎见宁片中冬青素A的含量测定。
Abstract:
Objective: To establish a HPLC method for the determination of the active component of ilexgenin A in Yanjianning tablets, and to investigate the content of ilexgenin A in different batches of Yanjianning tablets, so as to provide methodological basis for the quality control of Yanjianning tablets. Methods: Supersil ODS2 C18 (4.6mm × 200mm, 5 μm) chromatographic column was used; mobile phase acetonitrile (A)-0.1% phosphoric acid (B), gradient elution; column temperature: 25℃; Flow rate 1 mL·min-1; the detection wavelength was 210 nm, the sample size was 20 μL, and the content of ilocyanin A in Yanjianning tablets was determined by HPLC. Specificity, precision, stability, repeatability, detection limit, limit of quantification and recovery rate of addition were investigated to verify the scientific nature of the method. Results: Ilexgenin A had good linearity (r≥0.9999) in the range of 2.7 to 108 μg·mL-1. The specific property experiment showed that ilicin A in Yanjianning tablets had specific property. Precision RSD=1.00%; stability RSD=4.24%; reproducibility RSD=0.77%; the average recovery was 90.3%, RSDS=7.3% (n=9). Conclusion: The method is sensitive, accurate, reliable and has a good separation, which can be used for the determination of ilexgenin A in Yanjianning tablets.
