摘要:
目的:基于网络药理学研究香砂六君子汤治疗功能性消化不良的作用机制。方法:通过中药系统药理学分析平台(TCMSP)筛选出香砂六君子汤成分中口服生物利用度(OB)≥20%,药物相似度(DL)≥0.10的化合物作为药物有效成分,并获取其蛋白靶点信息,通过Gene Cards数据库筛选功能性消化不良靶点基因,利用Venny 2.1得到药物与疾病交集基因,利用Cytoscape 3.9.1构建“有效成分-靶点”网络及“药物-疾病-靶点”网络,使用String 11.5数据库构建PPI网络,并利用Matescape网站进行KEGG通路富集分析及GO功能富集分析。结果:香砂六君子汤治疗功能性消化不良的有效成分包括槲皮素、川陈皮素、柚皮素等,交集基因包括STAT3、VEGFA、IL6、IL1β、PTGS2、TNF等171个,参与细胞活性调节、细胞及信号通路凋亡调节、蛋白酶激活与结合、DNA转录等生物进程,主要参与的信号通路有AGE-RAGE信号通路、PI3K-Akt通路、NF-κB通路、JAK-STAT通路、cGMP-PKG通路等。结论:香砂六君子汤治疗功能性消化不良的作用机制可能与调节胃肠动力、抑制胃肠道炎症反应、调控交感神经敏感性等有关。
Abstract:
Objective: To study the mechanism of Xiangsha Liujunzi decoction in the treatment of functional dyspepsia based on network pharmacology. Methods: The compounds with oral bioavailability (OB)≥20% and drug similarity (DL)≥0.10 in Xiangsha Liujunzi decoction were selected as the active components of the drug through the Traditional Chinese Medicine System Pharmacology Analysis Platform (TCMSP), and the protein target information was obtained. The functional dyspepsia target genes were screened by Gene Cards database. and the drug disease intersection genes were obtained by Venny 2.1, The "active-ingredient-target" network and the "drug-disease-target" network were constructed by using Cytoscape 3.9.1, the PPI network was constructed by using String 11.5 database, and KEGG pathway enrichment analysis and GO function enrichment analysis were carried out by using Matescape website. Results: The effective components of Xiangsha Liujunzi decoction in the treatment of functional dyspepsia included quercetin, tangerine and naringin, etc., and 171 intersecting genes included STAT3, VEGFA, IL6, IL1β, PTGS2 and TNF etc., which participate in biological processes such as cell activity regulation, cell and signal pathway apoptosis regulation, protease activation and binding, and DNA transcription. The main signal pathways involved are AGE-RAGE signaling pathway, PI3K Akt pathway, NF-κB pathway, JAK-STAT pathway, cGMP PKG pathway, etc. Conclusion: The mechanism of Xiangsha Liujunzi decoction in the treatment of functional dyspepsia may be related to the regulation of gastrointestinal motility, inhibition of gastrointestinal inflammatory reaction and the regulation of sympathetic sensitivity.
