摘要:
目的:通过对大鼠子宫病理切片的观察以及检测子宫组织中tPA,PAI-1的表达,探讨壮药白花九里明提取物在纤溶系统方面的止血机制。方法:对健康的成年或未妊娠雌性老鼠构建不完整子宫流产模型,然后随机分成5组,每组10只,依次是白花九里明提取物低用量,白花九里明提取物中用量,白花九里明提取物高剂量,空白,模型,咖啡酸组6组。取子宫组织。进行组织病理切片的观察,并用免疫组化的方法测定子宫组织中tPA,PAI-1的表达。结果:tPA组经过方差齐性检验,P=0.092>0.05,方差齐。使用单因素方差分析进行两两比较,与正常组比较,模型组和和白花九里明低剂量组tPA水平均显著增加,P<0.05。与模型组比较,咖啡酸组、白花九里明低剂量组、白花九里明高剂量组tPA水平显著降低,P<0.05。白花九里明低剂量组和高剂量组之间比较,高剂量组较低剂量组tPA水平显著降低,P<0.01。PAI-1组经过方差齐性检验,P=0.056>0.05,表明方差齐。与正常组比较,白花九里明低剂量组PAI-1水平均显著增加,P<0.05。与模型组比较比较,咖啡酸组,白花九里明低剂量组、高剂量组PAI-1水平均显著增加,P<0.05。结论:白花九里明提取物具有促进子宫止血的作用,其机制可能与调节子宫纤溶系统相关。
Abstract:
Objective: To investigate the hemostatic mechanism of Zhuang medicine Blumea megacephala (Randeria) Chang et tseng extract in fibrinolytic system by observing the pathological sections of rat uterus and detecting the expression of tPAand PAI-1 in uterine tissue. Methods: An incomplete uterine abortion model was established in healthy adult or non pregnant female mice, and then randomly divided into five groups, with ten mice in each group. The order was 6 group of low dosage of Blumea megacephala (Randeria) Chang et tseng extract, medium dosage of Blumea megacephala (Randeria) Chang et tseng, high dosage of Blumea megacephala (Randeria) Chang et tseng extract, blank, model and caffeic acid group. Uterine tissue was taken. The expression of tPA and PAI-1 in uterine tissue was determined by immunohistochemistry. Results: TPA was tested by homogeneity of variance, P=0.092>0.05, and the variance was homogeneous. One way variance was used for pairwise comparison. Compared with the normal group, the TPA levels of the model group and the low-dose group of Blumea megacephala (Randeria) Chang et tseng were significantly increased, P<0.05. Compared with the model group, tPA levels in caffeic acid group, low-dose group of Blumea megacephala (Randeria) Chang et tseng and high-dose group of Blumea megacephala (Randeria) Chang et tseng were significantly decreased, P<0.05. Compared with low-dose group and high-dose group of Blumea megacephala (Randeria) Chang et tseng, tPA level in high-dose group was significantly lower than that in low-dose group, P<0.01. PAI-1 group passed the homogeneity test of variance,P=0.056>0.05,and the variance was uniform. Compared with the normal group, PAI-1 levels in the low-dose group of Blumea megacephala (Randeria) Chang et tseng was increased significantly, P<0.05. Compared with the model group,PAI-1 levels in caffeic acid group,low-dose group and high-dose group of Blumea megacephala (Randeria) Chang et tseng increased significantly, P<0.05. Conclusion: The extract of Blumea megacephala (Randeria) Chang et tseng has the effect of promoting uterine hemostasis, and its mechanism may be related to the regulation of uterine fibrinolytic system.
