目的:验证红霉素通过抑制RBBP7活性改善COPD小鼠糖皮质激素抵抗。方法:使用候选基因法查找与HDAC2、GR相关蛋白RBBP7,使用烟草烟雾暴露建立COPD小鼠模型,HE染色观察红霉素对COPD小鼠肺气肿的影响,rt-PCR验证红霉素对COPD小鼠肺组织中RBBP7、HDAC2、GR基因表达的影响, Westernblot研究红霉素对COPD小鼠肺组织中RBBP7、HDAC2、GR蛋白表达的影响。结果:蛋白质互作网络显示视网膜母细胞瘤结合蛋白7(retinoblastoma binding protein 7,RBBP7)与HDAC2、GR存在互作关系。红霉素可以改善烟草烟雾引起的肺气肿。COPD小鼠肺组织中RBBP7基因表达升高,HDAC2、GR基因表达减少。红霉素可以减少COPD小鼠肺组织中RBBP7基因表达,增加HDAC2、GR基因表达。同时,COPD小鼠肺组织中RBBP7蛋白表达增加,HDAC2、GR、P-GR蛋白表达减少,GR磷酸化水平降低,红霉素可以逆转上述趋势。结论:红霉素可以通过减少COPD小鼠肺组织中RBBP7活性,从而增加HDAC2、GR活性,最终改善COPD小鼠糖皮质激素抵抗。
Abstract:
Objective: To verify that erythromycin improves glucocorticoid resistance in COPD mice by inhibiting RBBP7 activity. Methods: the candidate gene method was used to find the protein RBBP7 related to HDAC2 and GR, and the COPD mouse model was established by tobacco smoke exposure. HE staining was used to observe the effect of erythromycin on emphysema in COPD mice. RT-PCR was used to verify the effect of erythromycin on the gene expression of rbbp7, HDAC2 and GR in the lung tissue of COPD mice. Western blot was used to study the effect of erythromycin on RBBP7, HDAC2 and GR in the lung tissue of COPD mice Effect of GR protein expression. Results: The protein interaction network showed that retinoblastoma binding protein 7 (RBBP7) interacted with HDAC2 and GR. Erythromycin can improve emphysema caused by tobacco smoke. The expression of RBBP7 gene increased and the expression of HDAC2 and GR gene decreased in the lung tissue of COPD mice. Erythromycin can reduce the expression of RBBP7 gene and increase the expression of HDAC2 and GR gene in the lung tissue of COPD mice. At the same time, the expression of RBBP7 protein increased, the expression of HDAC2, GR and P-GR Protein decreased, and the level of GR phosphorylation decreased in the lung tissue of COPD mice. Erythromycin can reverse the above trend. Conclusion: Erythromycin can increase the activities of HDAC2 and GR by reducing the activity of RBBP7 in the lung tissue of COPD mice, and finally improve the glucocorticoid resistance of COPD mice.
