摘要:
目的:探讨中药复方益肺温阳化浊汤对Aβ诱导的AD大鼠模型海马Aβ异常沉积与神经元凋亡的关系及Caspase-9、Caspase-3、Bcl-2、Bax 凋亡相关因子的影响。方法:采用 MorriS 水迷宫测试各组大鼠行为学,流式细胞仪检测神经细胞凋亡率,Western Blot 检测各组 Caspase-9、Caspase-3、Bcl-2、Bax 的表达;观察益肺温阳化浊汤低、中、高剂量对 Aβ 诱导的 AD 大鼠海马区 Aβ 含量及凋亡因子的影响。结果:模型大鼠经益肺温阳化浊汤处理后,海马区 Aβ 含量明显降低,神经元凋亡减轻,抑制 Caspase-9、Caspase-3、Bax 表达,上调抗凋亡蛋白 Bcl-2 的表达,并呈一定的剂量依赖性。结论:益肺温阳化浊汤可能通过促进 Aβ 诱导的 AD 大鼠海马区神经细胞中 Aβ 的清除,减轻 Aβ 的神经毒性,从而抑制神经元的凋亡,起到保护神经元的作用。
Abstract:
Objective: To investigate the relationship between Aβ abnormal deposition and neuronal apoptosis in Aβ-induced modelhippocampus and the effects of TCM compound Yifeiwenyanghuazhou decoction on apoptosis-related factors of Caspase-9 Caspase-3,Bcl-2 and Bax apoptosis. Methods: MothriS water maze was used to test the behavior of each group of rats, flow cytometry was used todetect the apoptosis of nerve cells, and Western Blot detected the expression of Caspase-9, Caspase-3, Bcl-2, and Bax in each group; Toobserve the effect of low, medium and high dose of Yifeiwenyang chemical turbid decoction on the Aβ content and apoptosis factor in thehippocampus of AD rats induced by Aβ. Results: After the treatment of Yiyangwenyangzhuo decoction, the content of Aβ in thehippocampus was significantly reduced, the apoptosis of neurons was reduced, the expression of Caspase-9, Caspase-3, Bax was inhibited,and the expression of the antiapoptosis protein Bcl-2 was increased, and showed a certain dose dependence. Conclusion:Yifeiwenyanghuazhuo decoction may reduce the neurotoxicity of Aβ by promoting the removal of Aβ in nerve cells in the hippocampus ofAD rats induced by Aβ, reduce the neurotoxicity of a β, thus inhibit the apoptosis of neurons, and play a role in protecting neurons.
