摘要:
建立高效液相色谱法快速测定多种维生素多种矿物质片中维生素 D 3 的含量,并采用加标回收率试验进行方法确认。样品在 0.2%的氨水中用碱性蛋白酶破坏维生素 D 3 的包埋物质,用乙醇浸提,正己烷萃取游离出维生素 D 3 。供试品溶液用正相液相色谱法净化,反相液相色谱法分离。维生素 D 3 在 0.0498μg/mL~0.9963μg/mL 浓度范围内线性关系良好(r=0.9999),平均加标回收率为 97.78%,RSD 为 3.26%,精密度和稳定性良好,RSD 分别为 2.69%和 1.58%。该方法准确、快速,可作为多种维生素多种矿物质片中维生素 D 3 含量的测定方法。
Abstract:
A rapid method for the determination of vitamin D 3 in multiple vitamins and minerals tablet by high performance liquidchromatography was established. The method was confirmed by standard-added recovery test. In 0.2% ammonia water, the encapsulatedsubstance of vitamin D 3 was destroyed by alkaline protease. Vitamin D 3 was extracted by ethanol and n-hexane. The sample solution waspurified by normal phase liquid chromatography and separated by reversed phase liquid chromatography. The linear relationship of vitaminD 3 was good in the concentration range of 0.0498μg/mL to 0.9963μg/mL (r=0.9999). The average recovery of standard addition was97.78%, RSD was 3.26%. The precision and stability of vitamin D 3 were good, RSD were 2.69% and 1.58%, respectively. The method isaccurate and rapid, and can be used for the determination of vitamin D3 in multiple vitamins and minerals tablet.
