摘要:
为完善毛鸡骨草种子质量检验方法,对其种子的生活力测定方法——四唑染色法进行了研究。通过考察种子预湿时间及方法、种子染色前处理、染色液浓度、染色温度、染色时间,确定了毛鸡骨草种子四唑染色条件。毛鸡骨草种子四唑染色方法为净种子经破除硬实后,于 30℃恒温水浸种 5h~6h 预湿,剥皮,1.0%TTC 溶液 30℃黑暗染色 5h~6h。结果表明,该方法对不同毛鸡骨草种子的染色率与发芽试验结果基本一致。毛鸡骨草种子的四唑测定稳定可靠,可代替发芽试验。
Abstract:
To improve the methods of seeds quality test of Abrus mollis Hance, the technique of triphenyl tetrazolium chloride (TTC)test was studied. The conditions of TTC test for Abrus mollis seeds were determined by investigating the way of wetting and pretreatment,the concentration of TTC solution, the dyeing temperature and time. The TTC test method of Abrus mollis Hance seeds are following: Afterthe hard seeds were broken, the seeds were soaked in constant temperature water at 30°C for 5h-6h, and then peeled. The seeds were dyed in1.0% TTC solution at 30°C for 5h-6h in darkness. The results showed that the dyeing rate of different Abrus mollis seeds was basically thesame as that of germination test. The TTC test of Abrus mollis Hance seeds is reliable, which can replace the germination test.
