条斑紫菜作为重要的经济海洋藻类,已在基因工程领域中做了诸多研究,包括核遗传转化和质体遗传转化,并且已经在紫菜中实现稳定表达和瞬时表达。文章研究了条斑紫菜对潮霉素的敏感性,获得筛选条件。结果表明,潮霉素对条斑紫菜叶状体的筛选浓度为2mg/mL,其中RZ 品系筛选时间为15 天;野生型品系为14 天。对条斑紫菜原生质体的筛选浓度为0.5mg/mL,其中RZ 品系筛选时间为7 天;野生型品系为6 天。该结果有助于基因编辑系统CRISPR/Cas9 技术在条斑紫菜研究中的应用。
Abstract:
Pyropia yezoensis is one of the most economically important marine species of the world. Many researches about it have been done in genetic engineering including establishing the platforms of nuclear transformation platform and plastid transformation platform and realize stable and transient expression in Pyropia yezoensis. The sensitivity of the gametophyte cells of Pyropia yezoensis to Hygromycin is 2.0 mg/mL, all cells of RZ died after 15 days while all cells of wild species died after 14 days; and that of protoplasts is 0.5 mg/mL, all cells of RZ died after 7 days while all cells of wild species died after 6 days. It is helpful for the application of CRISPR/Cas9 geno-editing systems in Pyropia yezoensis.
